The phase 3 trial assessed the safety and efficacy of niraparib plus abiraterone acetate (Zytiga) and prednisone compared with abiraterone acetate and prednisone plus placebo in patients with metastatic prostate cancer.

Similar to patients with metastatic prostate cancer harboring 

 mutations, the addition of niraparib to abiraterone acetate and prednisone elicited efficacy benefits in patients with 

 alterations, Sandhu explains. Notably, these benefits were seen across the study’s end points for these alterations, Sandhu adds.

Though efficacy benefits were observed in patients with 

alterations, they were not evident for all efficacy end points in these subgroups, Sandhu concludes.

The MAGNITUDE trial assessed abiraterone acetate (Zytiga) and prednisone with or without niraparib (Zejula) in patients with metastatic prostate cancer.

Prior to enrollment, patients were screened for genetic alterations, and patients were placed in homologous recombination repair (HRR)–positive and HRR-negative cohorts, Sandhu adds. After being assigned to these cohorts, patients were then randomized to receive either study treatment.

The primary end point of the clinical trial was radiographic progression-free survival by blinded central review. Secondary end points included cytotoxic chemotherapy, time to symptomatic progression, time to PSA progression, and overall survival, Sandhu concludes.

The phase 3 trial is assessing the safety and efficacy of niraparib (Zejula) in combination with abiraterone acetate (Zytiga) plus prednisone compared with abiraterone acetate plus prednisone and placebo in patients with metastatic prostate cancer.

The activity and safety of niraparib and abiraterone as single agents is well defined, and the adverse effects associated with the combination of these agents has proven manageable, Sandhu explains. In the phase 3 MAGNITUDE trial, no new safety signals arose with the combination, Sandhu adds.

At the 2022 ASCO Annual Meeting, additional data was presented from the trial, showing that quality of life was maintained with the niraparib regimen, Sandhu concludes.
